I first <\/span><span class=\"cf0\">worked<\/span> <span class=\"cf0\">under<\/span><span class=\"cf0\"> the supervision of <strong>Dr. <\/strong><\/span><strong><span class=\"cf0\">David-Alexandre<\/span> <span class=\"cf0\">Tr\u00e9gou\u00ebt<\/span><\/strong> <span class=\"cf0\">during<\/span> <span class=\"cf0\">my<\/span> <span class=\"cf0\">Master\u2019s<\/span> <span class=\"cf0\">internship<\/span><span class=\"cf0\">, <\/span><span class=\"cf0\">which<\/span> <span class=\"cf0\">focused<\/span><span class=\"cf0\"> on <\/span><strong><span class=\"cf0\">extending<\/span><span class=\"cf0\"> a miRNA <\/span><span class=\"cf0\">alignment<\/span><span class=\"cf0\"> pipeline to <\/span><span class=\"cf0\">other<\/span> <span class=\"cf0\">small<\/span><span class=\"cf0\"> non-<\/span><span class=\"cf0\">coding<\/span> <span class=\"cf0\">RNAs<\/span><\/strong><span class=\"cf0\">. <\/span><\/p>\n<p class=\"pf0\"><span class=\"cf0\">I <\/span><span class=\"cf0\">then<\/span> <span class=\"cf0\">pursued<\/span> <span class=\"cf0\">my<\/span><strong><span class=\"cf0\"> PhD <\/span><\/strong><span class=\"cf0\">under<\/span><span class=\"cf0\"> the supervision of <strong>Prof. St\u00e9phanie <\/strong><\/span><strong><span class=\"cf0\">Debette<\/span><\/strong><span class=\"cf0\"> as part of the <a href=\"https:\/\/rhu-shiva.com\/en\/\" target=\"_blank\" rel=\"noopener\">RHU SHIVA <\/a><\/span><span class=\"cf0\">project<\/span><span class=\"cf0\">, <\/span><span class=\"cf0\">aiming<\/span><span class=\"cf0\"> to <\/span><strong><span class=\"cf0\">characterize<\/span><span class=\"cf0\"> the <\/span><span class=\"cf0\">molecular<\/span> <span class=\"cf0\">mechanisms<\/span><span class=\"cf0\"> of <\/span><a href=\"https:\/\/vbhi-institute.org\/en\/vascular-brain-diseases\/\"><span class=\"cf0\">cerebral<\/span> <span class=\"cf0\">small<\/span> <span class=\"cf0\">vessel<\/span> <span class=\"cf0\">disease<\/span><\/a> <span class=\"cf0\">using<\/span><span class=\"cf0\"> multi-<\/span><span class=\"cf0\">omics<\/span> <span class=\"cf0\">approaches<\/span><\/strong><span class=\"cf0\">. <\/span><\/p>\n<p class=\"pf0\"><span class=\"cf0\">I <\/span><span class=\"cf0\">am<\/span> <span class=\"cf0\">now<\/span> <span class=\"cf0\">continuing<\/span> <span class=\"cf0\">my<\/span> <span class=\"cf0\">work<\/span><span class=\"cf0\"> as a <strong>postdoctoral <\/strong><\/span><strong><span class=\"cf0\">researcher<\/span><\/strong> <span class=\"cf0\">with<\/span><strong><span class=\"cf0\"> Prof. St\u00e9phanie <\/span><span class=\"cf0\">Debette<\/span><\/strong><span class=\"cf0\">, <\/span><span class=\"cf0\">mainly<\/span> <span class=\"cf0\">studying<\/span><strong><span class=\"cf0\"> the <\/span><span class=\"cf0\">proteomic<\/span><span class=\"cf0\"> and <\/span><span class=\"cf0\">epigenomic<\/span><span class=\"cf0\"> aspects of <\/span><span class=\"cf0\">cerebral<\/span> <span class=\"cf0\">small<\/span> <span class=\"cf0\">vessel<\/span> <span class=\"cf0\">disease<\/span><\/strong><span class=\"cf0\">.<\/span><\/p>\n<p class=\"pf0\"><span class=\"cf0\">*<\/span><span class=\"cf0\">Omics<\/span> <span class=\"cf0\">encompass<\/span><span class=\"cf0\"> a set of disciplines <\/span><span class=\"cf0\">that<\/span> <span class=\"cf0\">study<\/span><span class=\"cf0\"> all the components of a <\/span><span class=\"cf0\">biological<\/span><span class=\"cf0\"> system, <\/span><span class=\"cf0\">such<\/span><span class=\"cf0\"> as a <\/span><span class=\"cf0\">cell<\/span><span class=\"cf0\">, a tissue or an <\/span><span class=\"cf0\">organism<\/span><span class=\"cf0\">. <\/span><span class=\"cf0\">Instead<\/span><span class=\"cf0\"> of <\/span><span class=\"cf0\">analysing<\/span><span class=\"cf0\"> a single <\/span><span class=\"cf0\">gene<\/span><span class=\"cf0\"> or a single <\/span><span class=\"cf0\">protein<\/span><span class=\"cf0\">, <\/span><span class=\"cf0\">they<\/span><span class=\"cf0\"> examine the <\/span><span class=\"cf0\">entire<\/span><span class=\"cf0\"> network of components to <\/span><span class=\"cf0\">understand<\/span><span class=\"cf0\"> how <\/span><span class=\"cf0\">they<\/span> <span class=\"cf0\">function<\/span><span class=\"cf0\"> and <\/span><span class=\"cf0\">interact<\/span><span class=\"cf0\">. Multi-<\/span><span class=\"cf0\">omics<\/span><span class=\"cf0\"> data are <\/span><span class=\"cf0\">generated<\/span><span class=\"cf0\"> by <\/span><span class=\"cf0\">analysing<\/span> <span class=\"cf0\">several<\/span> <span class=\"cf0\">omics<\/span> <span class=\"cf0\">layers<\/span><span class=\"cf0\">, <\/span><span class=\"cf0\">such<\/span><span class=\"cf0\"> as <\/span><span class=\"cf0\">genomics<\/span><span class=\"cf0\">, <\/span><span class=\"cf0\">metabolomics<\/span><span class=\"cf0\">, transcriptomics and proteomics.<\/span><\/p>\n<h2 class=\"pf0\"><span class=\"cf0\">What did you investigate in this scientific publication?<\/span><\/h2>\n<p class=\"pf0\"><span class=\"cf0\">Our work focuses on <strong>cerebral small vessel disease<\/strong>. This is a complex condition, very common in the general population but still poorly understood, which contributes to many strokes and cognitive disorders, particularly in older adults. <\/span><\/p>\n<p class=\"pf0\"><span class=\"cf0\">It can be detected years before the onset of clinical symptoms, notably through brain imaging (MRI). <\/span><\/p>\n<p class=\"pf0\"><span class=\"cf0\">In this study, <strong>we examined proteins and associated genetic variants (proteogenomics) measured in the cerebrospinal fluid and blood, and their associations with MRI markers of cerebral small vessel disease, to identify biomarkers and better understand the underlying mechanisms of the condition<\/strong>. <\/span><\/p>\n<p class=\"pf0\"><span class=\"cf0\">The goal is to gain a detailed understanding of the pathways involved in the deterioration of small blood vessels in the brain and the resulting increased risk of stroke and dementia.<\/span><\/p>\n<h2 class=\"pf0\"><span class=\"cf0\">Why are you interested in this subject?<\/span><\/h2>\n<p class=\"pf0\"><span class=\"cf0\">We are interested in this topic because cerebral small vessel disease is a <strong>major public health issue<\/strong>: it affects a large number of people but is often diagnosed late. <\/span><\/p>\n<p class=\"pf0\"><span class=\"cf0\">By identifying earlier and more precise biomarkers, we hope <strong>to pave the way for better prevention and more timely diagnosis<\/strong>.<\/span><\/p>\n<h2 class=\"pf0\"><span class=\"cf0\">Why is this a major discovery?<\/span><\/h2>\n<p class=\"pf0\"><span class=\"cf0\">This is an important discovery because, until now, we have lacked reliable biomarkers to study and diagnose this disease. <\/span><\/p>\n<p class=\"pf0\"><span class=\"cf0\"><strong>Our approach combines, for the first time<\/strong> at this scale, <strong>proteomics and genomics in the two most informative fluids for the brain<\/strong>: <\/span><\/p>\n<ul>\n<li class=\"pf0\"><span class=\"cf0\">cerebrospinal fluid and<\/span><\/li>\n<li class=\"pf0\"><span class=\"cf0\">plasma. <\/span><\/li>\n<\/ul>\n<p class=\"pf0\"><span class=\"cf0\">This integrated analysis allows us <strong>to highlight key biological pathways<\/strong>, notably those related to inflammation, <\/span><span class=\"cf0\">already<\/span> <strong><span class=\"cf0\">from<\/span> <span class=\"cf0\">early<\/span> <span class=\"cf0\">adulthood<\/span><\/strong><span class=\"cf0\"> (~20 <\/span><span class=\"cf0\">years<\/span> <span class=\"cf0\">old<\/span><span class=\"cf0\">), and <\/span><span class=\"cf0\">extracellular<\/span><span class=\"cf0\"> matrix <\/span><span class=\"cf0\">dysfunction<\/span><span class=\"cf0\">. <\/span><\/p>\n<p class=\"pf0\"><span class=\"cf0\">Our <\/span><span class=\"cf0\">work<\/span> <span class=\"cf0\">also<\/span><span class=\"cf0\"> enables <strong>the identification of <\/strong><\/span><strong><span class=\"cf0\">proteins<\/span> <span class=\"cf0\">associated<\/span> <span class=\"cf0\">with<\/span><span class=\"cf0\"> the <\/span><span class=\"cf0\">risk<\/span><span class=\"cf0\"> of stroke and <\/span><span class=\"cf0\">dementia<\/span><\/strong><span class=\"cf0\">, and highlights <\/span><strong><span class=\"cf0\">potential<\/span> <span class=\"cf0\">opportunities<\/span><span class=\"cf0\"> for <\/span><span class=\"cf0\">drug<\/span> <span class=\"cf0\">repurposing<\/span><\/strong><span class=\"cf0\"> for <\/span><span class=\"cf0\">cerebral<\/span> <span class=\"cf0\">small<\/span> <span class=\"cf0\">vessel<\/span> <span class=\"cf0\">disease<\/span><span class=\"cf0\"> and <\/span><span class=\"cf0\">its<\/span> <span class=\"cf0\">clinical<\/span> <span class=\"cf0\">consequences<\/span><span class=\"cf0\">. <\/span><\/p>\n<p class=\"pf0\"><span class=\"cf0\">These<\/span> <span class=\"cf0\">findings<\/span> <span class=\"cf0\">therefore<\/span> <span class=\"cf0\">provide<\/span><strong><span class=\"cf0\"> a <\/span><span class=\"cf0\">true<\/span> <span class=\"cf0\">molecular<\/span> <span class=\"cf0\">fingerprint<\/span><span class=\"cf0\"> of the <\/span><span class=\"cf0\">disease<\/span><\/strong><span class=\"cf0\">, <\/span><span class=\"cf0\">which<\/span> <span class=\"cf0\">could<\/span> <span class=\"cf0\">transform<\/span><span class=\"cf0\"> the <\/span><span class=\"cf0\">way<\/span> <span class=\"cf0\">it<\/span> <span class=\"cf0\">is<\/span> <span class=\"cf0\">detected<\/span><span class=\"cf0\">, <\/span><span class=\"cf0\">monitored<\/span><span class=\"cf0\"> over time, and <\/span><span class=\"cf0\">even<\/span><span class=\"cf0\"> how future <\/span><span class=\"cf0\">treatments<\/span><span class=\"cf0\"> are <\/span><span class=\"cf0\">developed<\/span><span class=\"cf0\">.<\/span><\/p>\n<h2 class=\"pf0\"><span class=\"cf0\">What<\/span> <span class=\"cf0\">is<\/span><span class=\"cf0\"> the <\/span><span class=\"cf0\">next<\/span> <span class=\"cf0\">step<\/span><span class=\"cf0\">?<\/span><\/h2>\n<p class=\"pf0\"><span class=\"cf0\">The <\/span><span class=\"cf0\">next<\/span> <span class=\"cf0\">step<\/span> <span class=\"cf0\">is<\/span><strong><span class=\"cf0\"> to <\/span><span class=\"cf0\">validate<\/span> <span class=\"cf0\">these<\/span> <span class=\"cf0\">biomarkers<\/span><span class=\"cf0\"> in <\/span><span class=\"cf0\">larger<\/span><span class=\"cf0\">, longitudinal <\/span><span class=\"cf0\">cohorts<\/span><\/strong><span class=\"cf0\">.
